Have a look at the boot filesystem of a freshly imaged card that has not yet been booted. There should be a file named something like firstboot.sh (can't remember exactly), if you are using the official imager with pre-config. All the pre-config is in there, and you can add anything you need there. There are also the backend scripts in something like /usr/lib/rpi-sys-mods, which you can potentially hook into or manipulate from the first boot script.
