The first partition on (any) disk needs to be a 'fat' partition for the hardware to find necessary drivers, then it switches to a Linux ext4 partition to run the system.
Only if you're going to boot from that disc. If it's just for storage you don't need the FAT partition and probably shouldn't have it as it can cause confusion during boot.
